AstraINTERACTION

Does It Bind? Interaction Triage From Sequence

Folding a complex tells you a shape, not whether two partners actually interact. AstraINTERACTION scores the interaction directly from sequence — peptide and MHC, receptor and G-protein, kinase and substrate, and protein pairs — as a fast, confidence-gated pre-filter before you commit to structure prediction or the bench.

Immunopeptidomics-Grade on Peptide–MHC

On held-out peptide–MHC presentation, AstraINTERACTION reaches specialist accuracy — the read that matters for antigen presentation and immunogenicity triage — and extends the same engine across a family of interaction types.

What It Scores

One Engine, Many Interaction Types

Each interaction type is evaluated on a hard split — no sequence-similarity leakage between train and test — so the numbers reflect novel pairs, not memorized ones.

Held-out AUROC by interaction type
Peptide–MHC-I presentation0.98
Peptide–MHC-II presentation0.86
TCR–peptide (seen epitope)0.76
GPCR–G-protein coupling0.76
Kinase–substrate0.75
Protein–protein (general)0.69

AUROC on protein-disjoint / declustered splits. Local-motif interactions (peptide–MHC, kinase site) score highest; whole-protein pairs are coarser but still useful as a pre-filter.

How to Read It

A Confidence-Gated Pre-Filter

Cheap-Score Many Pairs

Screen a large set of candidate interactions from sequence before spending compute on folding or a wet-lab assay.

Keep the Survivors

Use it as a triage layer — carry the high-confidence interactions forward, drop the rest, and narrow the panel early.

It Abstains

The engine reports its own confidence and holds back when the signal is weak, rather than forcing a call it can't stand behind.

Antibody–antigen · exploratory

Antibody–antigen interface prediction runs through a structure-based cascade and is offered as confidence-gated, exploratory triage — surfaced only where the interface confidence is high, and flagged as predicted-and-unverified everywhere else. We report it as a research capability, not a mapped epitope.

Performance

Matches Specialists, Unified in One Engine

The value is one interaction engine that spans presentation, coupling, and enzyme–substrate reads — each evaluated honestly on hard splits — rather than a separate tool per interaction type.
